چکیده

Introduction: Chondrocytes within articular cartilage are surrounded by a complex pericellular microenvironment which, together with the cell, is termed the chondron. The mechanical and physicochemical properties of the chondron matrix will undoubtedly influence the metabolism of the chondrocytes and their response to joint loading. The aim of the present study was to examine cell morphology in chondrons isolated using two different methods and visualised in both unstrained and compressed agarose constructs. This methodology was combined with immunolocalisation of matrix components to provide insight into the mechanical integrity of the chondrons.

The protective role of the pericellular matrix in chondrocyte apoptosis.

INTRODUCTION This study was designed to quantify the role of the pericellular matrix (PCM) in chondrocyte apoptosis using chondrons, which are a cartilage functional unit including a chondrocyte and its associated PCM. METHODS Chondrocytes and chondrons were enzymatically isolated from human articular cartilage and exposed to monosodium iodoacetate (MIA) and staurosporine for apoptosis induct...

Chondrons from articular cartilage. V. Immunohistochemical evaluation of type VI collagen organisation in isolated chondrons by light, confocal and electron microscopy.

The pericellular microenvironment around articular cartilage chondrocytes must play a key role in regulating the interaction between the cell and its extracellular matrix. The potential contribution of type VI collagen to this interaction was investigated in this study using isolated canine tibial chondrons embedded in agarose monolayers. The immunohistochemical distribution of an anti-type VI ...

Biomechanical properties of single chondrocytes and chondrons determined by micromanipulation and finite-element modelling.

A chondrocyte and its surrounding pericellular matrix (PCM) are defined as a chondron. Single chondrocytes and chondrons isolated from bovine articular cartilage were compressed by micromanipulation between two parallel surfaces in order to investigate their biomechanical properties and to discover the mechanical significance of the PCM. The force imposed on the cells was measured directly duri...

Study of chondrogenic potential of stem cells in co-culture with chondrons

Objective(s): Three-dimensional biomimetic scaffolds have widespread applications in biomedical tissue engineering due to similarity of their nanofibrous architecture to native extracellular matrix. Co-culture system has stimulatory effect on chondrogenesis of adult mesenchymal stem cells. This work presents a co-culture strategy using human articular chondrons and adipose-derived stem cells (A...

Regional Variations in Growth Plate Chondrocyte Deformation as Predicted By Three-Dimensional Multi-Scale Simulations

The physis, or growth plate, is a complex disc-shaped cartilage structure that is responsible for longitudinal bone growth. In this study, a multi-scale computational approach was undertaken to better understand how physiological loads are experienced by chondrocytes embedded inside chondrons when subjected to moderate strain under instantaneous compressive loading of the growth plate. Models o...
